    Visualizing Case Studies

    Case studies contain valuable information about development records. They are, however, usually presented as textual documents. Here we begin an initial design for viewing case study data. A prototype for viewing case study data is created using information visualization techniques for the purpose of testing against existing techniques. The evaluations show that the new design is just as usable as existing techniques, making it suitable for future development. Future work may show our design can accomplish tasks both faster and more efficiently

    Coronary artery disease (CAD) is a leading global cause of morbidity and mortality, and improvements in the diagnosis and treatment of CAD can reduce the health and economic burden of this condition. Fractional flow reserve (FFR) is an evidence-based diagnostic test of the physiological significance of a coronary artery stenosis. Fractional flow reserve is a pressure-derived index of the maximal achievable myocardial blood flow in the presence of an epicardial coronary stenosis as a ratio to maximum achievable flow if that artery were normal. When compared with standard angiography-guided management, FFR disclosure is impactful on the decision for revascularization and clinical outcomes. In this article, we review recent developments with FFR in patients with stable CAD and recent myocardial infarction. Specifically, we review novel developments in our understanding of CAD pathophysiology, diagnostic applications, prognostic studies, clinical trials, and clinical guidelines

    Automated reduction of submillimetre single-dish heterodyne data from the James Clerk Maxwell Telescope using ORAC-DR

    With the advent of modern multi-detector heterodyne instruments that can result in observations generating thousands of spectra per minute it is no longer feasible to reduce these data as individual spectra. We describe the automated data reduction procedure used to generate baselined data cubes from heterodyne data obtained at the James Clerk Maxwell Telescope. The system can automatically detect baseline regions in spectra and automatically determine regridding parameters, all without input from a user. Additionally it can detect and remove spectra suffering from transient interference effects or anomalous baselines. The pipeline is written as a set of recipes using the ORAC-DR pipeline environment with the algorithmic code using Starlink software packages and infrastructure. The algorithms presented here can be applied to other heterodyne array instruments and have been applied to data from historical JCMT heterodyne instrumentation.Comment: 18 pages, 13 figures, submitted to Monthly Notices of the Royal Astronomical Societ

    Actical Accelerometry Cut-points for Quantifying Levels of Exertion: Comparing Normal and Overweight Adults

Weight, body fatness and ambulatory pattern all have the potential to affect accelerometer output and cause differences in output between overweight and normal-weight adults. The purpose of this study was to determine if Actical (Philips Respironics, Bend, OR) activity count cut-points for moderate and vigorous intensity exercise are different for overweight adults compared to normal-weight adults. Overweight adults with BMI \u3e25 kg/mĀ² (n=29) and Normal-Weight adults (n=25) walked at 3.2 and 4.8 kmāˆ™h-1 and ran at 6.4 kmāˆ™h-1 on a treadmill while simultaneously wearing an Actical accelerometer and obtaining measurements of oxygen uptake. Counts per minute (countsāˆ™min-1) were determined at 3 METS (moderate) and 6 METS (vigorous) using ROC curves. The countsāˆ™min-1 at 3 METs was 1726 and 1923 countsāˆ™min-1 for Overweight and Normal-Weight groups, respectively. The cut-points at 6 METs were 4117 and 4032 countsāˆ™min-1 for Overweight and Normal-Weight groups, respectively. The differences between groups were not statistically significant (p\u3e0.73 for both). Correlations between BMI and countsāˆ™min-1 were not significant (p\u3e0.05) at any speed for the Normal-Weight group but were significant at 3.2 and 4.8 kmāˆ™h-1 for the Overweight group. Although there appears to be some relationship between activity countsāˆ™min-1 and BMI, the results suggest that similar cut-points may be used for normal weight and overweight adults. However, the greater variability in counts at each speed and lower ROC curve areas for overweight adults suggest that it is harder to classify the activity intensity of overweight subjects compared to normal weight subjects

    BackgroundNumerous preclinical studies have supported the theory that enhanced activation of mGluR5 signaling, due to the absence or reduction of the FMR1 protein, contributes to cognitive and behavioral deficits in patients with fragile X syndrome (FXS). However multiple phase 2 controlled trials in patients with FXS have failed to demonstrate efficacy of compounds that negatively modulate mGluR5, including two phase 2b randomized controlled trials (RCT) of mavoglurant (AFQ056, Novartis Pharma AG), when the primary measures of interest were behavioral ratings. This has cast some doubt onto the translation of the mGluR5 theory from animal models to humans with the disorder.MethodsWe evaluated social gaze behavior-a key phenotypic feature of the disorder-and sympathetic nervous system influence on pupil size using a previously-validated eye tracking paradigm as a biobehavioral probe, in 57 adolescent or adult patients with FXS at baseline and following three months of blinded treatment with one of three doses of mavoglurant or placebo, within the context of the AFQ056 RCTs.ResultsPatients with FXS treated with mavoglurant demonstrated increased total absolute looking time and number of fixations to the eye region while viewing human faces relative to baseline, and compared to those treated with placebo. In addition, patients had greater pupil reactivity to faces relative to baseline following mavoglurant treatment compared to placebo.DiscussionThe study shows that negative modulation of mGluR5 activity improves eye gaze behavior and alters sympathetically-driven reactivity to faces in patients with FXS, providing preliminary evidence of this drug's impact on behavior in humans with the disorder

    Recognition of nectin-2 by the natural killer cell receptor T cell immunoglobulin and ITIM domain (TIGIT)

    T cell immunoglobulin and ITIM domain (TIGIT) is an inhibitory receptor expressed on the surface of natural killer (NK) cells. TIGIT recognizes nectin and nectin-like adhesion molecules and thus plays a critical role in the innate immune response to malignant transformation. Although the TIGIT nectin-like protein-5 (necl-5) interaction is well understood, how TIGIT engages nectin-2, a receptor that is broadly over-expressed in breast and ovarian cancer, remains unknown. Here, we show that TIGIT bound to the immunoglobulin domain of nectin-2 that is most distal from the membrane with an affinity of 6 Ī¼m, which was moderately lower than the affinity observed for the TIGIT/necl-5 interaction (3.2 Ī¼m). The TIGIT/nectin-2 binding disrupted pre-assembled nectin-2 oligomers, suggesting that receptor-ligand and ligand-ligand associations are mutually exclusive events. Indeed, the crystal structure of TIGIT bound to the first immunoglobulin domain of nectin-2 indicated that the receptor and ligand dock using the same molecular surface and a conserved ā€œlock and keyā€ binding motifs previously observed to mediate nectin/nectin homotypic interactions as well as TIGIT/necl-5 recognition. Using a mutagenesis approach, we dissected the energetic basis for the TIGIT/nectin-2 interaction and revealed that an ā€œaromatic keyā€ of nectin-2 is critical for this interaction, whereas variations in the lock were tolerated. Moreover, we found that the C-Cā€² loop of the ligand dictates the TIGIT binding hierarchy. Altogether, these findings broaden our understanding of nectin/nectin receptor interactions and have implications for better understanding the molecular basis for autoimmune disease and cancer

    Lifestyle-Related Determinants of Obesity Among Omani Children

    Objectives: This study aimed to examine the relationship between body mass index (BMI) z-scores and lifestyle-related factors including nutrition, physical activity, screen time and time spent sleeping in 6ā€“10-year-old Omani children. Methods: This cross-sectional study included mother-child dyads that were recruited from five provinces in Oman. Childrenā€™s BMI measurements and questionnaires on nutrition intake, physical activity, screen time and time spent sleeping and a single-day dietary recall were collected. Results: A total of 197 dyads were included in this study. The children's mean age was 7.7Ā Ā± 1.6 years and 53% were female. In this study, 17.4% of the children were classified as overweight or obese. No significant relationship was found between the children's BMI z-scores and nutrition intake, moderate-to-vigorous physical activity time or screen time (P ā‰„0.05). Increased time spent sleeping at night was positively associated with childhood obesity (P <0.05). Conclusion: The relationship between obesity and caloric intake, physical activity and screen time among children younger than 10 years seems to be moderated by certain factors that need to be investigated.
